    Minor Amendments made to AgResearch GM Cattle Approvals

    The Environmental Risk Management Authority (ERMA New Zealand) has made three minor amendments to approvals held by AgResearch to carry out research using genetically modified cattle.

    The first minor amendment is to the field test of genetically modified cattle to research the effects of increasing the casein content of milk. This approval (GMF98009 Part 1) has been amended to extend the time frame by another three years to provide more time for the research objectives to be achieved.

    The second amendment is an approval to enable research into genetically modified cattle to express functional therapeutic proteins in their milk and to study gene function and genetic performance. This approval (GMD02028) is broad as it covers the development of a widely specified range of genetically modified cattle. It has been amended to include the use of a single selection marker gene (conferring resistance to the antibiotic puromycin) which is frequently used in research of this type. Puromycin is not an antibiotic used in human or animal medicine.

    The third amendment is also to approval GMD02028. This change clarifies that AgResearch is able to use genetically modified cattle embryos or semen previously developed by partner organisations. The genetic modifications of these cattle embryos and semen are limited to the specified range of modifications originally approved by the Authority. The change has also clarified that AgResearch are able to further develop this material into live offspring by using common animal husbandry techniques. This change does not alter the level of risk or uncertainty involved in the project neither does it change the parameters in the approval for the cattle.

    The ability to make amendments to existing approvals is covered by Section 67A of the Hazardous Substances and New Organisms Act. This section of the Act enables the Authority to make amendments to existing approvals that are minor in effect or correct a minor or technical error. They do not alter either the purpose of the approval or the level of risk.

    "One amendment allows AgResearch to continue with their existing field test of genetically modified cattle for a further three years. The decision to allow the extension took into consideration cultural concerns of local iwi and will require on going consultation with Ngati Wairere” said Rob Forlong, Chief Executive of ERMA New Zealand. “The other two changes are to AgResearch’s development approval so they can use imported transgenic embryos and sperm to investigate the concept of cattle producing human lactoferrin in their milk. They will not, however be able to produce a commercial herd on the basis of this decision."

    "It is important to emphasise that these cattle will remain in strict containment and be subject to on-going controls enforced by the Ministry of Agriculture and Forestry."

    Blijkbaar moet er nog een aparte toestemming komen voor commerciele productie van lactoferrin.

    ...dat was toch wel de bedoeling dacht ik.

    Blijkbaar moet er nog een aparte toestemming komen voor commerciele productie van lactoferrin.
    De ERMA moet zich ook indekken, ik interpreteer het zo dat ze mogen beginnen met het opbouwen van de kudde, dat duurt zomaar 3 jaar, dan een evaluatie en dan inderdaad het vervolg indien de evaluatie positief uitvalt. Juist in die 3 jaar moet AgResearch aantonen dat er géén nadelige gevolgen voor het milieu enz. vast zitten aan deze wijze van rhLF produceren. Positieve ontwikkeling denk ik.

    Drie jaar wachten op start producitie van rhLF is niets mis mee, denk ik.

    AgResearch is de bedoeling gaat rhLF (Lactoferrine) produceren voor Pharming. AgResaerch heeft op dit moment de beschikking over ingevoerde embryo’s en sperma. Dit “materiaal” is genetisch aangepast i.v.m. het produceren van rhLF.
    Als het goed is hebben ze nu een aantal koeien geselecteerd. Een deel van de koeien zal een embryo ontvangen en een ander deel sperma. De draagtijd van een koe is 9 maanden. Niet alle koeien kunnen gelijk behandeld worden, even wachten tot de eisprong ( ze moeten “tochtig” zijn). Niet alle embryo’s zullen uitgroeien en niet alle inseminaties zullen meteen succesvol zijn.

    De “embryo”-kalfjes in wording zijn al geselecteerd op geslacht: vrouwenlijk, want die kunnen melk gaan geven. Van de andere kalveren die uit een inseminatie voortkomen, zal ca 25% rhLF kunnen produceren: volgens de wetten van Mendel is ca 50% nakomelingen vrouwelijk en daarvan ook 50% zal het rhLF gen meekrijgen. Dat is een statistisch gegeven, dat kan in de praktijk mee of tegenvallen: meer of minder vrouwelijke kalveren, meer of minder mét rhLF-gen.
    Een kalf geeft nog geen melk. Wordt als 1-jarige geïnsemineerd, dat moet anders gaan ze geen melk geven. Dan ben je al weer een jaar verder, voordat je de eerste melk hebt.

    Intussen worden alle effecten naar de omgeving beschreven en gerapporteerd. Als alles dan OK is, kan er toestemming gevraagd worden om de productie daadwerkelijk te gaan opstarten.

    Als bevestiging van zaken die we al wisten een nuttig bericht.
    Het laat o.m. het tijdperspectief zien waarin we moeten denken vòòrdat rhLF commercieel interessant wordt.Met drie jaartjes wachten daar is niks mis mee,zoals Gert terecht opmerkt.
    Positief voor forumhangouderen is ook dat zij voorlopig niet bang hoeven te zijn voor hun favoriete tijdverdrijf. En analisten zijn dus niet dom in alles wat zij schrijven.

    Gelukkig staan dr al een paar koeien in Wisconcin en gooien ze de melk daar niet door het putje.

    Pharming’s recombinant hLF approach
    Pharming has developed transgenic cattle for the production of rhLF in milk. A production method that fits nutraceutical development, as cow’s milk is a common food source worldwide. Pharming also purifies recombinant hLF from the milk for pharmaceutical applications. It is available in two grades; one for oral applications and one of higher purity that can be administered intravenously.

    Pharming’s human lactoferrin is stable in aqueous solutions and therefore well suited for the treatment of dry eyes and eye infections. And the transgenic cattle can produce it in sufficient quantities: a herd of only 20 cows can supply the entire amount needed for this indication in the US.

    zie je wel dat iedereen het over koeien heeft, daarin wijkt SNS dus niet af.
    runderen is wel heel erg chique voor zo'n poep-blubberend beest, waar natuurlijk ook gewoon "melk" uitkomt.

    Als je Googelt kom je wel artikelen tegen over de opening van Vienna Farms in Wisconsin, maar berichten van na die tijd heb ik niet gevonden. Ook geen berichten over productie. Nogmaals: zie wat Pharming op de eigen site hier over zegt.

    Zonder nadere bronnen houd ik het er op, dat men daar het basismateriaal (embryo's en sperma) maakt dat Agresearch nu gebruikt. Met o.a. de technologie die Pharming gekocht heeft van ProBio is men steeds beter in staat om embryo's met het rhLF gen te maken. Stier Herman was een toevalstreffer, men is denk ik nu steeds beter in staat om gericht bepaalde eiwitten toe te voegen aan embryo's. In de toekomst niet alleen voor rhLF, maar ook voor andere eiwitten.

    the MAF Biosecurity Authority/ERMA New Zealand Animal Health and Welfare Standard
    154.03.06 Containment Standard for Field Testing of Farm Animals, and/or any requirements of the
    relevant Animal Ethics Committee (AEC), and should at all times be the minimum number of
    animals required to obtain statistically significant results. In any event, the total number of cattle
    involved in the field test shall not at any one time exceed 200 animals.

    De gekloonde koeien zijn door Pharming en het Amerikaanse biotechnologische bedrijf Infigen gecreëerd om medicinale melk te produceren. Werknemers van Pharming verzorgen de dieren bij DeForest in de Amerikaanse staat Wisconsin.

    Correct me if I'm wrong:

    zowel Marcel Wijma (zie draadje van Devali The Tide Is Turning) als First Berlin (Zie rapport, te vinden op Pharming.nu) voorzien commerciële verkoop van rhLF in 2006.
    Waar Wijma dat op baseert weet ik niet, FB baseert het op "uitlatingen van het managment".
    In het rapport van FB staat netjes waar de productiefaciliteit staat voor C1 Inhibitor, namelijk bij Diosynth. Nergens staat waar de productiefaciliteit staat voor rhLF, wel wordt gesproken over partners enz.

    De berichten uit ca 2001 van Devali: prima, maar dat is echt oud nieuws. Een half jaar later werden de contracten opgezegd ivm de financiële perikelen van Pharming en werd beslag gelegd op de koeien van Pharming...... Dat is ook weer achter de rug.

    Er is echt nog geen productiefaciliteit voor rhLF, laat maar zien dan. Commerciële verkoop van rhLF in 2006? Hoedat?

    het lijkt erop dat je gelijk hebt :)

    Pharming has developed transgenic cattle that produce rhLF in their milk.
    Lactoferrin has anti-infective and probiotic activities that are beneficial for people suffering from Gastro-Intestinal (GI) problems. Pharming has developed protein purification protocols to produce two purity grades. One grade for oral applications and one grade for i.v. applications. Pharming wants to introduce the oral grade rhLF into the Japanese market to be formulated in foods that will improve GI health. At this moment Pharming is building a production herd that will deliver the first milk in 2006 and will be in full production in 2007. At this moment a GRAS Notification filing is prepared for the US-FDA and Pharming expects to be granted a GRAS notification in 2006.
    Small amounts of material are available for testing.
