    JNJ-3989

    JNJ-3989, formerly ARO-HBV, is being developed in collaboration with Janssen Pharmaceuticals, Inc. to be a potentially curative therapy for patients with chronic hepatitis B infection, when used in combination with other direct antivirals. JNJ-3989 is a sub-cutaneous, ribonucleic acid interference (RNAi) therapy candidate which is designed to silence all HBV gene products and intervenes upstream of the reverse transcription process where current standard-of-care nucleotide and nucleoside analogues act. The company believes this may allow the body’s natural immune defenses to clear the virus and potentially lead to a functional cure.

    Disease

    Chronic hepatitis B viral infection, caused by HBV, is a potentially fatal liver disease Current drugs suppress viral replication but rarely lead to a functional cure and therefore must be taken indefinitely. Developing curative therapy is a priority.

    There are approximately 300 million people worldwide. In the immune tolerant phase of chronic infection, which can last for many years, the infected person typically produces very high levels of viral DNA and viral antigens. However, the infection is not cytotoxic and the carrier may have no symptoms of illness. Over time, the ongoing production of viral antigens causes inflammation and necrosis, leading to cirrhosis and liver cancer (HCC). Hepatitis B is responsible for 80% of primary liver cancers.

    The current standard of care for treatment of chronic HBV infection is a daily oral dose of nucleotide/nucleoside analogs (NUCs) or a regimen of interferon injections for approximately one year. NUCs are generally well tolerated, but patients may need lifetime treatment because viral replication often rebounds upon cessation of treatment. Treatment with pegylated interferon for up to 2 years can lead to functional cure in up to 10% of treated patients, however this regime is poorly tolerated and not widely used.

    Data are now available (from Coldlander, on twitter) that show sustained knockdown of S-antigen after only 3 treatments with JNJ3989...the e+ patient results are particularly impressive.

    From the HBV Endpoints Treatment Workshop (AASLD/EASL): "A complete sterilizing cure, i.e., viral eradication from the host, is unlikely to be feasible. Instead, a functional cure characterized by sustained loss of hepatitis B surface antigen with or without hepatitis B surface antibody seroconversion, which is associated with improved clinical outcomes, in a higher proportion of patients than is currently achieved with existing treatments is a feasible goal."

    JNJ will be presenting data later this month...expect more details about how JNJ-3989 will be used as a key component of cHBV treatments to achieve functional cures.

    Gisteren stuk Arrowhead verkocht en Nio bijgekocht. Ik denk da ik vrijdag niet ga afwachten. Na Galapagos, Kiadis, Pharnext, Genfit, Quantum Genomics heb ik het beetje gehad met biotech. Mijn verlies van Gala al voor de helft goedgemaakt met Nio en de rest komt ook wel. Veel succes nog iedereen maar ik kap met Biotech.

    Jammer, ik ben ook onderuit gegaan met Kiadis, voor mij nu vooral Biotech met een bredere pipe-line. (en cash natuurlijk).
    Dus dan is A een logische keuze.

    Nio maar ook Nikola zijn de afgelopen tijd heel hard gestegen. Ik ben van mening dat deze aandelen nog meer High risk is dan Arrowhead. Bij Arrowhead is het nu een kwestie van geduld. De auto aandelen zullen fors naar beneden glijden als er in de herfst wel een flinke aandelen dip komt. Misschien heb ik ongelijk ;-) Ik blijf maar van deze gehypte aandelen af. Bio in de porto is al risico genoeg.....

    Ik ga beide Arrowhead en NIO in voor de long ! We zullen zien wat er gebeurt ermee!
    Arrowhead zie ik ook zeker goed stijgen, maar ik ben nog maar een noobie dus mij moet je zeker niet volgen hahaha

    Ik blijf rustig zitten, dit scenario zien we we meer vlak voor goed nieuws, bepaalde partijen proberen de kleine belegger schrik aan te jagen en proberen stop-losses te laten afgaan om zelf goedkoop te kunnen bijkopen ...
